Immunodetection methods are used to identify specific proteins in cells, tissue and in analytical techniques like immunoblotting. This technique requires the use of gold labeled immunodetection reagents, of which Aurion offers an extensive, ready-to-use range. We can, however, also provide you with an in-depth explanation of the required conjugation protocol and methods, enabling you to manufacture gold nanoparticle conjugated ligands and immunodetection reagents by yourself.

Immunodetection protocol methods for immunogold and/or immunogold silver staining

Main steps in the methods of the immunodetection protocol are blocking, washing, primary antibody incubation, immunogold conjugate incubation and if appropriate, silver enhancement. Immunodetection is possible using post-embedding or pre-embedding methods, each requiring a specific incubation protocol. In general, a two-step immunodetection protocol is used. As ultra-small gold conjugates require a different approach to conventional-sized conjugates, the two immunodetection protocols are described in-depth in our ‘Incubation protocols’ section.
