Immunohistochemistry protocol

Following the correct steps in the immunohistochemistry protocol is important to obtain optimal signal to noise ratios. Aurion has developed special reagents for this purpose, more specifically BSA-c and R-Gent SE-LM. This reagent develops the signal obtained with the immunogold staining protocol to make it visible in bright field light microscopy. We provide researchers with an in-depth explanation of the proper protocols in light microscopy for pre- and post-embedding immunogold labeling.

Post-embedding and pre-embedding immunogold silver staining technique

In immunohistochemistry, the immunogold silver enhancement and staining technique is a good alternative for methods using enzyme markers. A permanent signal will be generated when using ultra-small gold conjugates, BSA-c buffer additive and R-Gent SE-LM. The signal can be combined with general counter-staining methods. To fully benefit from the advantages of the immunogold silver staining method, we offer high-quality, ready-to-use ultra-small gold conjugated antibodies suited for the immunohistochemistry labeling protocol. We offer an in-depth overview of the necessary incubation steps in our ‘Incubation protocols’ section. Here, you can find the correct protocols for both conventional immunogold reagents and ultra-small reagents for light microscopy.
